Nile red: a selective fluorescent stain for intracellular lipid droplets
We report that the dye nile red, 9-diethylamino-5H-benzo[alpha]phenoxazine-5-one, is an excellent vital stain for the detection of intracellular lipid droplets by fluorescence microscopy and flow cytofluorometry. The specificity of the dye for lipid droplets was assessed on cultured aortic smooth muscle cells and on cultured peritoneal macrophages that were incubated with acetylated low density...
متن کاملEnergy-transfer measurements on a double fluorescent labeled ribonuclease A.
Two fluorescent groups have been covalently attached to ribonuclease A: first, the alpha-amino group is labeled upon reaction with fluorescein isothiocyanate, and second, one of the active site histidine residues is modified by N-[[(iodoacetyl)amino]ethyl]-5-naphthylamine-1-sulfonic acid. Among the products of these two successive chemical modifications, a derivative bearing one label on Lys-1 ...
متن کاملIntracellular click reaction with a fluorescent chemical Ca2+ indicator to prolong its cytosolic retention.
The powerful strategy of "intracellular click reaction" was used to retain a chemical Ca(2+) indicator in the cytosol. Specifically, a novel clickable Ca(2+) indicator "N3-fura-2 AM" was coupled with dibenzylcyclooctyl-modified biomacromolecules via copper-free click reaction in living cells and Ca(2+) oscillation was observed for an extended period of time.
متن کاملribosomal dna intracellular journey and the new upcoming ribosomal organelle
human mitochondrial dna (mtdna) is 16,569 base pairs (bp) in length, coding for 37 genes. during the course of evolution, nearly all the genes expressing ribosomal proteins and ribosomal rnas (rrna) genes have been transferred from the mitochondria to the nucleus. however, mitochondrial dna contains two ribosomal rnas genes (12s and 16s), which have not yet transferred to the nucleus. these two...

Optical techniques have revolutionized the investigation of cardiac cellular physiology and advanced our understanding of basic mechanisms of electrical activity, calcium homeostasis, and metabolism. Although optical methods are widely accepted and have been at the forefront of scientific discoveries, they have been primarily applied at cellular and subcellular levels and considerably less to w...
